dc.description.abstract Sets of experiments were conducted in the Genetic lin2ineering Laboratory. ('vtogenetic Department, Bangladesh Jute Research Institute (BJRI ). DhaLa during the period from March 2005 to May 2006 to lid fill the objectives of the study. One of the major constraints of getting plant regeneralion from the explants of C'. o/UOP1OILV was the production of healthy seedlings iii vitro. Seeds of C'. o/iloriouc germinated on both agar supported hormone free MS medium and cotton supported hormone free liquid MS medium. Ihe percentage of seeds germinated on cotton supported medium was found to be iiiuch higher than seeds germinated on agar supported medium. i'his system ensured maximum of an average of 97.55% seed geriflinatioll. Whereas, in case of agar supported medium an average of 45.8% seeds could be germinated with very slow growth. The seedling grown on cotton supported medium were found to much more healthy than seedlings growli on agar supported medium. Plant regeneration obtained from die cotyledonary petioles of the varieties of C'. olitorinus on MS agar solidi fled medium supplemented by IAA 0.5 mg/I and I3AP 3.0 mg/I. Plant regeneration was also observed in dil'lerent level of ph and dillerent concentration of FeSO4 and it was found that 5.5 and 56 mg/I gave highest result. Ihe efficiency of plant regeneration from the cotton supported seedlings was found to be better as the agar supported seedlings. The plantlets produced roots on hormone free MS mediuni rapidly. After translèr to soil the plants grew into maturity and produced fruits. No morphological changes were noticed. en_US
