dc.description.abstract Incidence of potato virus Y in the three selected potato growing areas of Bangladesh, viz. Savar, Manikgonj and Munshiganj were investigated from field samples. Overall, three upazilla (sub-station) from each district (which were at least 20 km apart) were selected for samples collection and disease incidence monitoring. Collected samples were storage at -20°C and analyzed in Molecular Biology and Plant Virology lab, Central Research Laboratory of Sher-e-Bangla Agricultural University (SAU), Dhaka. In the all investigated areas, PVY appeared in severe to moderate level and their incidence was 317%. The sources of potato seed tubers were studied and observed that in most of the cases farmers are continuously cultivating same field and used their own seed tubers that was kept in cold storage condition. The frequency of the storage at cold storage was 67.6% which was the highest among the all sources. In this study, PVY was detected through reverse transcriptase polymerase chain reaction (RT-PCR). A 480 bp amplicon of PVY- coat protein (CP) gene was amplified and the nucleotide sequences was also amplified in this study. en_US
