Bottle gourd (Lagenaria siceraria Mol.) is one of the most important Cucurbitaceous&#13;
vegetables in the world. It is widely grown for leafy and green fleshy vegetables. This study&#13;
was conducted to determine the molecular diversity among eight bottle gourd hybrid varieties&#13;
which were collected from three renowned private Seed Companies (Metal Agro Limited, ACI&#13;
Limited, Lal Teer Seed Limited) of Bangladesh. A total of nine primers (RAPD, ISSR and&#13;
SSR) were used to determine polymorphism among bottle gourd hybrids to produce scorable&#13;
DNA bands. Total 26 bands were amplified by nine primers.  Among them 18 were&#13;
polymorphic bands. The range of DNA amplification varied from 1000-100 bp. Average&#13;
64.82% polymorphism was obtained among the genotypes. Genetic diversity ranged from&#13;
0.2188 to 0.5000 and the frequency of major allele ranged from 0.5000 to 0.8750. Nei’s genetic&#13;
distance ranged from 0.2308 to 0.7308. The PIC value ranged from 0.1948 to 0.3750 with an&#13;
average value 0.3243. The PIC value indicates that the studied bottle gourd hybrids had&#13;
medium level of molecular diversity. A dendrogram indicating the relative genetic similarity of&#13;
the bottle gourd hybrids was constructed which followed three major clusters (A, B and C)&#13;
among the studied material. The two hybrid H7 and H8 showed more similarity in genetic&#13;
identity that fall in the same group-B which has collected from same seed company. Likewise,&#13;
the hybrid H5 and H6 classified in the same group-C were also obtained from same seed&#13;
company. Group-A contains the two hybrids H3 and H4 which are collected from Lal Teer&#13;
Seed Ltd. The present investigation revealed that, the bottle gourd hybrid developed by the seed&#13;
companies has more similarity within their own genotypes but to some extend genet ically&#13;
diverse between different seed companies. These experimental findings can be used for the&#13;
protection of hybrids in commercial purpose and in managing bottle gourd genetic resource in&#13;
Bangladesh.

The&#13;
healthy,&#13;
disease&#13;
free&#13;
shoot&#13;
tips&#13;
of&#13;
0.501.0&#13;

cm length were used as explants, It was sterilized by 0.1% HgCl&#13;
2&#13;
 mixing&#13;
with few drops Tween-20. The explant was inoculated in MS media&#13;
supplemented with different combination of the BA (1.0, 2.0, 3.0, 4.0 and 5.0 mg/l)&#13;
and IBA (0.50, 1.00, 1.50, 2.00 and 2.50 mg/l) either alone or in combination.  The&#13;
maximum days for shoot induction were recorded in control treatment. The&#13;
highest number of shoots (3.60) in BA 2.0 mg/l and leaves (9.60) per explant&#13;
observed in BA 3.0mg/l. In contrast combine treatment of 2.0 mg/l BA+1.00&#13;
mg/l IBA performed best in case of percentage shoot induction (80%) and&#13;
number of shoots (3.60).The highest length of shoots (3.66 cm) in BA 2.0 mg/l+&#13;
IBA 2.0mg/l . It was noticed that the minimum days to shoot induction, percent&#13;
shoot initiation and no. of shoot were recorded in 2.0 mg/l BA+1.0 mg/l IBA.&#13;
The highest number of leaves per explant was recorded in 2.0 mg/l BA+0.5&#13;
mg/l IBA. The highest number of root was observed in 2.0 mg/l BA+ 1.5 mg/l&#13;
IBA and the highest percent root initiation in 3.0 mg/l BA+1.5  mg/l IBA &#13;
Overall, micropropagation was found to be very effective and promising method&#13;
in the proliferation of strawberry and this experiment can be a useful tool for&#13;
breeding program of strawberry. Finally, the survival rate of micropropagated&#13;
plantlets were observed in open atmosphere 75% and 80% in controlled&#13;
condition.

February&#13;
2018&#13;
to&#13;
June&#13;
2018&#13;
to&#13;
evaluate&#13;
the&#13;
effect&#13;
of&#13;
different&#13;
&#13;
plant&#13;
&#13;
growth regulators (BA, IBA) on in vitro plantlet regeneration of Turmeric. The&#13;
healthy, disease free sprouts of turmeric were used as explants which were inoculated in&#13;
MS media supplemented with different conc. of Benzyl adenine (BA) (1.0, 2.0, 3.0 and 4.0&#13;
mg/L) and IBA (1.0, 2.0, 3.0 and 4.0 mg/L) either alone or in combination for shoot and&#13;
root regeneration. The experiments were arranged in Randomized Completely Block&#13;
Design (RCBD) with three replications. The maximum shoot induction was obtained with&#13;
2.0 mg/L BA. The highest shoot length (5.93 cm) was noticed in BA 2.0 mg/L + IBA 3.0&#13;
mg/L and minimum shoot length (1.4 cm) with control MS dose was observed. The highest&#13;
percentage (81.00%) of shoot induction was recorded with BA 2.0 mg/L in minimum (14.0&#13;
days). Where, IBA 3.0 mg/L had produced the highest percentage of root induction&#13;
(82.00%) required minimum (20 days). The highest length of shoot (3.4 cm, 5 cm and 5.9&#13;
cm) at 21, 28 and 42 DAI respectively was found in BA 2.0 mg/L + IBA 3.0 mg/L. The&#13;
treatment BA 2.0 mg/L + IBA 1.0 mg/L gave the highest number of root (6.0, 8.6, 12.67)&#13;
at 21, 28 and 42 DAI respectively and the highest length of root (2.7 cm, 4.8 cm and 5.4&#13;
cm) at 21, 28 and 42 DAI, respectively was found in BA 2.0 mg/L + IBA 1.0 mg/L.&#13;
Regenerated plantlets showed,  100% survival during in growth chamber conditions and&#13;
86.66% in shade house stage of hardening and 83% in open atmosphere at direct sunlight.&#13;
Therefore, an efficient protocol has been developed for in vitro propagation of Turmeric&#13;
which has great commercial value for year round production in Bangladesh.

Rice (Oryza sativa L.) is one of the most important cereal crop around the world. It is the&#13;
staple food in Bangladesh. This study was conducted in Biotechnology Laboratory of the&#13;
Department of Biotechnology, Sher-e-Bangla Agricultural University, Sher-e-Bangla Nagar,&#13;
Dhaka-1207 from the period of November 2018 to April 2019 to determine the molecular&#13;
diversity among four rice hybrid varieties using Random Amplified Polymorphic DNA&#13;
(RAPD) and Single Sequence Repeat (SSR) markers. A total of 21 RAPD primers and 4 SSR&#13;
primers were used to determine polymorphism among the rice hybrids to produce scorable&#13;
DNA bands. Total 82 bands were amplified through RAPD primers. Among them 26 were&#13;
polymorphic bands. The range of DNA amplification varied from 1300-150 bp. The rate of&#13;
polymorphism was obtained about 30.33%. Lower level of polymorphisms in hybrids&#13;
indicated that there is a basic similarity among the varieties which is due to the common&#13;
ancestor and the similarity of the selective traits. Genetic diversity ranged from 0 to 0.5000&#13;
and the frequency of major allele ranged from 0.5000 to 1.0000. Nei’s genetic distance&#13;
ranged from 0.1190 to 0.2262. The PIC value ranged from 0 to 0.3750 with the average value&#13;
0.1052. The PIC value indicates that the studied rice hybrids had low molecular diversity. A&#13;
dendogram indicating the relative genetic similarity of the rice hybrids was constructed which&#13;
followed three major clusters (A, B and C) among the studied material. Two SSR primers&#13;
showed polymorphic band which can be used as an evidence of variety protection data. This&#13;
experimental findings can be used for the protection of hybrids in commercial purpose and in&#13;
managing rice genetic resource in Bangladesh.
